On November 4, 2026, the Union Health Ministry proposed amendments to Schedule M of the Drugs Rules, 1945, to revise microbial contamination limits in Grade A areas of sterile pharmaceutical manufacturing facilities.
Proposed microbial monitoring changes
The proposal seeks to align Indian requirements with the World Health Organization’s Technical Report Series No. 1044, Annexure 2, on Good Manufacturing Practices for Sterile Pharmaceutical Products. It would tighten the Grade A limit from less than 1 colony-forming unit (CFU) to “no growth” and require an investigation whenever microbial growth is detected. Gazette notification GSR No. 953(E), dated October 28, 2026, invited stakeholders and the public to submit objections and suggestions.
Important Facts for Exams
- CFU interpretation: A colony-forming unit count relies on culture and includes viable cells only; microscopy, by contrast, also counts dead cells.
- Environmental controls: GMP requires controlled manufacturing environments to prevent cross-contamination from adulterants and allergens that could make products unsafe.
- Process validation: GMP calls for validation of all critical processes to demonstrate consistency and compliance with specifications.
